Interspecies differences in cytochrome P450-mediated metabolism of neonicotinoids among cats, dogs, rats, and humans

چکیده

Neonicotinoid insecticides are used for agricultural and non-agricultural purposes worldwide. Pets directly exposed to neonicotinoids in veterinary products through environmental contamination. Cytochrome P450 (CYP) is among the most significant xenobiotic metabolizing enzymes that oxidizes several chemicals, including neonicotinoids. However, CYP activities metabolite compositions of neonicotinoid metabolites unknown domesticated pet species. Our objectives were reveal differences (imidacloprid, clothianidin, acetamiprid) common species (cats dogs), humans, rats. The results indicated CYP-mediated metabolism was different depending on each neonicotinoid. Among these four species, kinetics imidacloprid rats have highest rate oxidation 4OH-imidacloprid, while greatest enzyme 5OH-imidacloprid found humans. Clothianidin rapidly metabolized 1-methyl-3-nitroguanidine dm-clothianidin rats, but cats humans showed lowest formation dm-clothianidin. acetamiprid dm-acetamiprid N-acetyl-acetamiprid determined be significantly higher compared other further studies should targeted at identifying hepatic using recombinant enzymes.

Cytochrome P450 1A1/2 mediated metabolism of trans-stilbene in rats and humans.

It was demonstrated that trans-stilbene was metabolically activated to the estrogenic compound by rat liver microsomes (Sugihara et al., Toxicol. Appl. Pharmacol., 167, 46-54 (2000)). In this study, determination of the isoforms of cytochrome P450 involved in the oxidation of the proestrogen, trans-stilbene, to its hydroxylated metabolites was examined. When trans-stilbene was incubated with ra...

Cytochrome P450-mediated metabolism of vitamin D.

The vitamin D signal transduction system involves a series of cytochrome P450-containing sterol hydroxylases to generate and degrade the active hormone, 1α,25-dihydroxyvitamin D3, which serves as a ligand for the vitamin D receptor-mediated transcriptional gene expression described in companion articles in this review series. This review updates our current knowledge of the specific anabolic cy...

In vitro cytochrome P450-mediated metabolism of exemestane.

Exemestane is a potent and irreversible steroidal aromatase inhibitor drug used for the treatment of estrogen receptor-positive breast cancer. Our aim was to identify and assess the contribution of the specific cytochromes P450 (P450s) responsible for exemestane primary in vitro metabolism. With the use of high-performance liquid chromatography and liquid chromatography-tandem mass spectrometry...

Short-term fasting alters cytochrome P450-mediated drug metabolism in humans.

Experimental studies indicate that short-term fasting alters drug metabolism. However, the effects of short-term fasting on drug metabolism in humans need further investigation. Therefore, the aim of this study was to evaluate the effects of short-term fasting (36 h) on P450-mediated drug metabolism. In a randomized crossover study design, nine healthy subjects ingested a cocktail consisting of...

Kinetics and regulation of cytochrome P450-mediated etoposide metabolism.

Etoposide is a DNA topoisomerase II inhibitor widely used in the treatment of a variety of malignancies that is also associated with therapy-related leukemia. The cytochrome P450 (P450)-derived catechol and quinone metabolites of etoposide may be important in the damage to the MLL (mixed lineage leukemia) gene and other genes resulting in leukemia-associated chromosomal translocations. Kinetic ...
